What is established is an architectural oddity rather than an activity: in H37Rv every other cytoplasmic E-set domain is an accessory module inside a large enzyme (366-1215 aa: bglS, alpha-mannosidase, ceramidase, glgE, glgP, malQ, pepN), whereas here the Ig-like domain is the entire protein, and Pfam describes DUF6941 itself as a family of uncharacterised bacterial proteins sharing similarity with the Ig-like modules of xylosyltransferases, alkylpurine DNA glycosylases and cycloisomaltooligosaccharide glucanotransferases. The domain leaves no N-terminal room for a signal peptide, so the predicted cytoplasmic localisation is constrained by the architecture and not only by DeepTMHMM - which makes the nearest structural analogues found by Foldseek (RbmA, the Vibrio cholerae biofilm scaffolding protein; complement C3) informative as folds but misleading as compartments, both being extracellular adhesion/matrix Ig-folds. No paralogue carries DUF6941 elsewhere in H37Rv.
